Flow Cytometry Core Facility

Instrumentation
MoFlo- a 3-laser system (488nm, 457nm, 630nm) high-speed sorter MoFlo represents the most sophisticated state of the art instrumentation currently available on the market. Cell sorter can be configured in a variety of ways: Higher-speed (processing 25,000 cells per second), aseptic (sterile) in tubes (5-ml, 15ml, 50ml) or plates (24-well, 96-well), single-cell, four-way and multi-laser, multi-color (seven color) cell sorting. Cell sorting on the MoFlo is a highly complex task, and is typically performed by facility staff only. Consultation and discussion of any planned experiments are therefore indispensable.

-Immunophenotyping (up to 7-color) FITC, CFSE, GFP, YFP, PE, RFP, PI, PE-Cy5.5, PE-Cy7, Alexis 700, APC, APC-Cy7
-DNA cell cycle combined with intracellular antigens (PI only)
-Viability and Apoptosis assays -Cytokine expression -Fluorescent proteins(CFP, GFP, YFP, Tomato-RFP, RFP)

User Guidelines

In General
It is imperative that you plan your experiments carefully. The Core is always available to consult on protocols and procedures prior to implementation and we strongly recommend you to come to us before starting any new experiment so as to avoid any expensive mistakes. This includes confirming that the fluorescent labels and their combinations can be analyzed on the instrumentation the MoFlo.

Access Policy
The highest priority is given to Dr. Abbas and Dr. Bluestone and PPG, who participate in the organization of the Core and already included the services of the Core in their peer-reviewed funding. It has always been our goal to make service as accessible as possible for all users and the policies are continuously updated to fairly serve the entire research community.

On line schedule
Every month the 15th through 20th is Dr. Abbas and Dr. Bluestone and PPG labs pre booking time. The monthly schedule then opens to the public on the 21st each month for all other researchers to sign-up. If you would like to be noticed when the schedule opens, send an email to shuwei.jiang@ucsf.edu. Your email will be added to the mailing list.

Book Sort Time
The sign up a sorting slot is easy.
Go to Yahoo, choose Calendar.
User ID: moflohsw542
Password: sofast.
After typing in the user ID and PW, the today’s schedule will show up. To view the monthly schedule, click at the “month” on the top menu. Then click at an open slot, type in your name, lab phone#, and the time needed.

Rules
The signing up rules:
1. When sign up a sort, fill in your NAME, LAB, and PHONE #.
2. If the open block is more then what you need, please do not brake in the middle. Please try to use either the beginning or the end. Then post the rest time “open”.
3. Please do not block more time then you acutely need. You will be charged for the time you sign up.
4. Each lab should only sign up for 10 hours unless there is still open time available.

The cancellation rules:
1. When you cancel your sort, change your name to “OPEN”, so other users can fill in the slot.
2. A cancellation made ONE week ahead, no charge will be applied to your account.
3. If a cancellation is made within 5 days, no charge will be applied to your account if someone else takes the time.
4. A cancellation made within 48 hours, you will be charged in the full amount unless someone else fills in the slot.
5. When you cancel your sort, you should always send an email to shuwei.jiang@ucsf.edu, or call at 514-3138.

Acknowledgements
Any formal presentations or publications resulting from work performed in the Core should be acknowledged and a reprint should be provided. The following statement is suggested: "We would like to acknowledge the assistance of the Flow Cytometry Core Facility at the Department Pathology and Diabetes Center, UCSF.” Final Words Investigators are reminded to include the services of the Core in their grant applications and the core will be happy to assist in designing any future projects. This will ensure that the Core is an appropriate venue for the proposed research, that the project design incorporates the efficient use of the Facility, and that the costly resources of the Facility are used for the greatest scientific benefit.
